Znanstveniki so nedolgo tega odkrili nov »mini organ« oziroma organel. Tvorba, ki so jo poimenovali ekskluzom, je bila podrobneje predstavljena v prispevku, objavljenem v znanstveni reviji Molecular Biology of the Cell. Kot se zdi, je novoodkriti mini organ oziroma organel (predelek celic s specifično funkcijo) povezan z avtoimunskimi obolenji. Kot lahko preberemo v študiji, je notranjost ekskluzoma sestavljena iz plazmidov – majhnih, krožnih molekul DNK, ki so sposobne samostojne podvojitve (neodvisno od kromosomov).

Zanimiva sestava novoodkritega organela

Organeli so specializirane podenote celic, nekakšni celični organi, pri čemer ima vsak organel svojo, nepogrešljivo vlogo. Ekskluzom, najnovejši član družine organelov, je bil odkrit v celični plazmi. To je precej nenavadno, saj lahko pri večini celic, ki imajo jedro (gre za t. i. evkariontske celice), DNK najdemo prav jedru, pri čemer je DNK varno skrit v kromosomih. Kot omenjeno, je za novoodkriti organel značilna malce drugačna zgradba. Majhne krožne molekule DNK oziroma plazmidi, iz katerih je sestavljen, so sicer značilni predvsem za bakterije in druge mikroorganizme. Zanimiv pa je tudi izvor plazmidov, ki sestavljajo ekskluzom. Del teh plazmidov pride v celico od zunaj, izvor drugega dela plazmidov pa so telomeri – zaščitne »kapice«, ki jih je mogoče najti na koncu kromosomov. Telomerski obročki, ki so bili odkriti v ekskluzomu, so sicer zelo pogosti v rakavih celicah.

Kot se zdi, organel ščiti genetsko integriteto celice

Odkritje ekskluzoma nakazuje, da so celice sposobne zaznati tuj DNK in DNK, ki ni več uporaben, ter ga odstraniti iz jedra oziroma premakniti v celični plazmo, kjer se združi s plazmidi, ki prihajajo od zunaj. Na podlagi tega je mogoče sklepati, da ima novoodkriti mini organ oziroma organel zaščitno funkcijo, saj pomaga ohranjati genetsko celovitost celice. Sodeluje pri procesu, ki je del osnovne »celične higiene«. Plazmidni geni virusov in bakterij bi se lahko sicer prepisali v beljakovine, kar bi lahko negativno vplivalo na celično fiziologijo. Precej očitno je, da se po zaslugi ekskluzoma tveganje za to občutno zmanjša, vendar ni povsem jasno, katere dodatne naloge opravlja ta novoodkriti organel. Domneva se denimo, da je ekskluzom pomemben tudi za celični imunološki spomin, vendar gre za zdaj samo za hipotezo, ki jo bo treba preveriti z dodatnimi raziskavami.

Ekskluzom morda povezan z vnetnim odzivom pri avtoimunskih obolenjih

Znano je, da se v celični plazmi na DNK veže beljakovina, posledično pa se lahko začnejo sproščati snovi, ki spodbujajo vnetni odziv telesa. To lahko privede do aktivacije imunskega sistema. Če se ista beljakovina veže na plazmide, ki sestavljajo ekskluzom, lahko torej tudi to sproži provnetne signale. Ena izmed možnih posledic so avtoimunske bolezni, na primer sistemski eritematozni lupus, pri katerih začne telo napadati in uničevati lastne celice.

Za dokončne odgovore bodo potrebne dodatne raziskave

Pri primerjavi ekskluzoma in celičnega jedra lahko odkrijemo določene podobnosti. Za obe tvorbi je tako značilen DNK, ovit v membrano, kar je mogoče razumeti kot namig na evolucijski izvor celičnega jedra pri evkariontskih celicah. Ekskluzom je sicer precej preprostejši od celičnega jedra. Pri njem je mogoče opaziti določene značilnosti, ki so sicer tipične za zgodnje faze razvoja celičnega jedra. Tako je mogoče ekskluzom razumeti kot nekakšno poskusno verzijo oziroma predstopnjo jedra, iz katerega so sestavljene celice. Toda za to, da bo mogoče v polnosti razumeti, kako in zakaj se je ekskluzom razvil ter kakšno vlogo ima v celicah in pri razvoju različnih bolezni, posebno avtoimunskih obolenj, bo najverjetneje potrebnih še precej raziskav.
